Driving directions have been added to the Google Maps API v3. For version three of the API directions have been split into two, DirectionsService which passes directions queries and DirectionsRenderer which displays the results on the map.
Google have provided an explanation of the concepts involved, examples and a reference guide. Mapperz has also created a couple of examples; Directions and Directions with a Directions Panel.
